Pen tool and/or vector shapes, clipping mask, and most likely some layer effects.

You made it separate from the rest and removed the default block of white that would fill it around it. You made it seem like two images.

If you have squared edges like I used there you can just simply save it as a gif with transparency enabled. There is also a more complicated alpha channel transparency supported by the PNG filetype that I prefer because it will not distort your edges. I'll can try to find you a tutorial or I can write one I suppose.
