How long does someone have to reply to a post ie: a fight or challenge?
What happens if they miss that time?

Competitors have approximately one day to reply to a CHALLENGE, not a fight.

now, what happens if they don't reply. Just trying to stay on top of things, don't want to do this wrong.
So let's say that in my fight against Yamcha. Say Yamcha doesn't post for 24 hours, what happens? I already posted the first fight post, so if he doesn't post, he loses?

My post was intended specifically for a challenge, not for a fight itself.
As for a fight, I would encourage you not to fight someone who will be missing for days at a time. Starting a fight with someone right before they go out of town isn't the wisest thing to do.
